Alelos de histocompatibilidade de classe II associados ao perfil imunogenetico de individuos portadores de psoriase vulgar

RESUMO

In this thesis, we analyzed 100 healthy control individuals and 60 individuals with Psoriasis vulgaris ofthe population of Campinas, a city in the southeast of Brazil. Typification of class II HLA alleles (HLA-DRBI-5 e DQB1) was carried out through the DNA/PCR/SSP molecular method at medium and high resolution. DNA was extracted through a salting-out procedure of peripheral blood of each individual participating in the research. 13 DRB 1 alleles, 3 DRB3 alleles, 1 DRB4 allele, 2 DRB5 alleles, and 5 DQB1 alleles were quantified at a medium or generic resolution using the PCR/SSP technique for primers of "One Lambda"; and, 45 DRB1 alleles were quantified at a high resolution or specific resolution in analyzed patients. Results analysis showed positive associations suggesting susceptibility to Psoriasis vulgaris for HLA-DRB3*02 (p<0,05, X2=4,36, RR= 2,14) and HLA-DRB1 *0102 alleles (p<0,05, RR= 5,44). Negative associations indicating protection from Psoriasis vulgaris were found for HLA-DRB4*01 (p<0,001, X2= 6,55, RR= 0,53) and HLA-DRBl *1302 alleles (p<0,05, RR= 0,23). Analysis carried out in haplotypes revealed positive association for HLA-DRBl *01O2/DQBl*05 (p<0,05, RR= 5,44) and HLA-DRBl *0701/DQBl *03 alleles (p<0,02, RR=9,00). Findings permit to define examined control population as Miscellaneous Caucasoide and suggest a possible association ofthe DRB1 allele with the group of patients showing an early onset ofthe illness. However, data has to be confirmed in future studies
