Analise genetica da capacidade de regeneração de plantas "in vitro" em tomate

RESUMO

With the objective of studying the genetic control of in vitro plant regeneration ability in tomato, high and low expressing genotypes for that trait were initially identified, using a previously defined culture medium. For callus induction, the utilized medium was that of Murashige &, Skoog (MS), supplemented with IAA (1 mg/l) and BAP (0,5 mg/I) and BAP (2,25 mg/l)for plant regeneration IAA(0,5 mg/l)and BAP(0,5 mg/l) was added to the same MS medium.Hypocotyl segments were inoculated for 15 days in callus lnduction medium and thereafter, subcultured every 30 days in regeneration medium, for two months.. Subsequent1ycalli were transfered to MS medium without growth regulators for one month more, to promot_ bud elongation and plantlet growth. LYCODersicon pimpinellifolium, genotype WV-700, with high plant regeneration ability (92%.) used as polen danar, was crossed to 5 different recalcitrant b esculentum genatypes for that charactet, namely: VFN-S (5%),Petomech (7%.),Santa Rita (0%.), beyand two cerasif orme genotypesof L esculentum, namely CNPH-oSO (8%.) and Red Cherry (5%.). VFN-8 and Santa Rita cultlvars are used in salads or for consuption in natura , whereas Petomech is destinated for industry; cerasiforme genotypes have no economic importance. Statistical analysis were based upon frequency data of calli that regenerated plants (R) in F1, F2, reciprocal crosses and backcrosses, with all five genotypes that did not regenerate plants (NR). F1 hibrids showed high plant regeneration ability (75-90%.) with and effect of dominance; no differences were observed in recipocral crosses. Segregation ratio in F2 generation was near to 9:7(R:NR) and in backcrosses the ratio expressed was near to 1 : 3 (R:NR), both not signíficant at the 5%. level, in x2- Tests, suggesting an interaction of two dominant genes in the expression of the trait studied. These results indicate that the transfer of plant regeneration ability from wild tomato genotypes to commercial ones, must be relatively easy once a qualitatively inherited dominant trait involved
