Analise genetica da produção de lipidios de aspergillus nidulans

RESUMO

Three auxotrophics mutants of A.nidulans were crossed among themselves as result we obtained three diploids. From the diploid MSE // pro A1; paba A6; y A1 were obtained nine sectors by haploidization. The haploids strains were cultivated in a complete medium and in a minimum supplemented medium. The diploids were grown in a minimum and the segregates grew in a minimum supplemented medium. The lipid composition of the studied organisms was determined by gaseous chromatography. The instrument used was a CG mod 37-D cromatographer with ionization flame. The identifications were based on the retention time and relative percentage of grease acids was determined by the peak areas on the chromatograms. The differences found related to the medium can be observed in relation to miristic acid in the bi A1; meth G1 and MSE strains in complete medium. The palmitoleic occurs in the minimum supplement medium in the MSE and bi A1; meth G1. The insaturated acid of margaric only occurs in the haploids grown in complete medium. The C 20:2 acid occurs only in the pro A1; paba A6; y A1 and bi A1, meth G1 haploids in complet medium and in diploid MSE // bi A1; meth G1. Some acids remained constant in all strains studied independently from the medium. They are palmitoleic, linoleic and linolenic wich may indicate a genetic influence on the lipids composition of A.nidulans. A genetic interpretation of production of acids miristic and margaric is suggested
