Caracterização de genes análogos de resistência (RGAs) em cultivares de Musa acuminata contrastantes em resistência à estresses bióticos.

RESUMO

Many cultivars of bananas (Musa spp.) are triploid or sterile diploid and their evolution occurs only through somatic mutation. As a result, this culture may lack resistance to pests and diseases. Several resistance genes (R genes) have been characterized in plants, conferring resistance to bacteria, fungi, nematodes and viruses. The characterization of resistance gene analogues (RGAs) was held in Musa acuminata cultivars contrasting in resistance to Black Sigatoka disease caused by Mycosphaerella fijiensis. The work was directed to the families of R genes NBS-LRR and family of extracellular LRR transmembrane domain linked to a Ser / Thr quinase in the cytoplasm (RLK). Primers designed in previous work for NBS-LRR generated about 200 sequences organized into 38 contigs with similarity to RGAs in this class. The differentiation of contigs based on the genotypes was not observed, showing in the same contig sequences from both cultivars resistant to Black Sigatoka, as susceptible to it. The tests were also performed for products amplified by primers directed to RLK, generating 65 complete ORFs. There was a high percentage of clones sequenced that obtained similarity to two families from genes containing kinase (RLK and STK) and once again the differentiation of protein kinase contigs could not be made based on cultivars resistant and susceptible to Black Sigatoka . Phylogenetic analysis was carried out by maximum parsimony with bootstrap 1000 replicates, showing high divergence between the sequences of RGAs both NBS-LRR, RLK how much. The identification and cloning of R genes in Musa offer new potential for genetic improvement through marker-assisted selection and genetic modification.
