Comparação ultraestrutural entre folhas hidratadas e dessecadas de tres especies de Velloziaceae : duas revivescentes e uma sensivel a dessecação

RESUMO

Xerophyta plicata Spreng. (Velloziaceae) is a Brazilian desiccation tolerant Angiosperm that occurs in granitic rock outcrops from Rio de Jane1ro State to Rio Grande do Norte State. Although leaves do not fall during the drought periods, they become yellow, shrinked, folded and twisted. When water is available again, leaves distend and retake the green colour. Mesophyll cells survive the water inlet and outlet cycles by means of ultrastructural changes that are responsable for maintaining their viability. These changes are similar to limited degradative processes in which remnants of the vital structures are preserved. The use of a non-aqueous fixative solution for dry tissues prevent the occurrency of fixation artefacts in these that are very reactive to water. A step by step 9-day study of the desiccation process by Transmission Electron Microscopy recognizable (TEM) showed few changes in the nucleus and modified-chloroplasts even in totally dry leaves (relative water content (RWC) of 0%). Other organelles disappear or are not recognizable. Desiccated cells show a compact and central cytoplasm that maintains contact with the cell walls by a few thread-like projections. Cell walls are very folded. During rehydration, changes in shape and volume (studied by Scanning Electron Microscopy, SEM) occur along with morphological metabolical changes, in order to reorganize the cell ultrastructure. A step by step study on the first 2 days of rehydration fixatives was carried out by TEM with aqueous and non-aqueous fixatives. Resurrection occurs through a sequence of cellular events: swelling of the organelles, vacuolation of the cytoplasm, motion of the cytoplasm to the peripheral portion of the cell and reorganization of the internal structure of the cells?Note: The complete abstract is available with the full electronic digital thesis or dissertations
