RESUMO

Tissues of 36 cultivars of Phaseolus vulgaris L. wee established in vitro aiming toward the development of a protocol for plant differentiation from bean tissues in order to apply the in vitro techniques for the improvement of this species. Callus cultures of Phaseolus vulgaris cv. Moruna were established in the presence of MS medium supplemented with KIN and 2,4-D (1-50 uM). Callus prolifaration differed according with the source of explant (hypocotil, comtyledone, embryo, primary leaf or trifoliolate leaf). Root differentiatiosn was inhibited at levels of 2,4 - D 5 mu M or higher, in the presence of KIN. Leaf cultures were more favorable to callus induction that other sources of explants. Primary leaves of Moruna were inoculated to test different interaction of cytokinin (KIN, 6 - BA and 2ip = 0 - 75 mu M) versus auxin (2,4 - D = 0 - 50 mu M; NAA = 0 - 75 mu M; IAA = 0 - 100 mu M). The interaction of KIN/2,4 - D was the most effective for callus proliferation. In the presence of these growt regulators creamish friable callus developed with the absence of roots. A genotypic characterization among four - cultivars of P. vulgaris were obtained based on callus induction from trifoliolate in the pesence of KIN/2,4 - D.
