Detecção de periodontopatógenos em amostras de biofilme subgengival e em lesões vasculares (ateromas)

RESUMO

In last 25 years, the relation between cardiac diseases and periodontal disease has been extensively studied. Among the approaches used with this purpose, there is the detection of periodontopathogens in both, oral cavity and atheroma lesions. The aims of this study were: 1) determine the prevalence of Aggregatibacter actinomycetemcomitans and Porphyromonas gingivalis in the subgingival biofilm of patients with cardiac diseases and with different periodontal conditions; 2) determine the prevalence the prevalence of A. actinomycetemcomitans and P. gingivalis in atheromatous lesions biopsies. Previous to the cardiac surgery, patients were submitted to oral clinical evaluation and the subgingival biofilm were collected with curettes and suspended in TE (pH 7.6), the biopsies of atheromatous lesions were obtained during the surgery and stored in TE (pH 8.0) at -20C until processing. The DNA of both samples (biofilm and biopsies) were extracted and submitted to polymerase chain reaction with 16SrRNA species-specific primers in order to detect the species above. The results showed that, in oral biofilm, A. actinomycetemcomitans and P. gingivalis were detected in 3 (14,28%) and 10 (47,62%) out 21 patients respectively. In atheromatous lesions, P. gingivalis could be detected in 5 (23,8%) and A. actinomycetemcomitans in 3 (14,28%) of 21 biopsies. These data suggest that eventhough the atheromatous lesions can harborthe periodontopathogens (A. actinomycetemcomitans and P. gingivalis) they are not always found together in atheromatous lesions and oral biofilm.
