RESUMO

Insulin leads an anorexigenic response by acting on hypothalamic neurons. Studies suggest that resistance to its action is involved with diet-induced obesity (diet rich in saturated fatty acids) and its association with hypothalamic inflammation. However, the effects of polyunsaturated fatty acids on insulin and inflammatory signaling pathways are poorly undestood. Thus, we evaluated the hypophagic effect of ICV insulin and molecular steps of insulin signaling and inflammatory response in the hypothalamus of rats fed Control diet (15% energy from fat) or high-fat diets (50% energy from fat) enriched with soy or fish oils from 2 until 4-months-old. Both groups had similar energy intake and protein levels of IR, IRS-1, IRS-2, Akt, mTOR, p70S6K, AMPK, TLR2, TLR4, IL-6R, TNF-R e NFÛBp50. Althought the animals fed the soy diet had normal body mass, lipid profile and peripheral insulin sensitivity, they had higher fat pads weight and serum leptin. Moreover, the soy rats were not sensitive to hypophagic effect of ICV insulin, regarding with the result that no significant phosphorylation of protein kinase B (PKB/Akt). When we assessed the hypothalamic inflammatory profile, we observed that soy diet lead to greater TRAF6 and NFÛBp65 expression and that fish diet reduced the levels of TNF-, IL-6 and TRAF6 and increased IL-10 receptor levels, an anti-inflammatory protein. Overall, these results suggest that, unlike the high-fat soy diet, the high-fat fish diet did not have deleterious effects on the central anorexigenic mechanism of insulin, probably due to improve of peripheral metabolism and hypothalamic inflammatory profile, suggesting a protection against obesity.
