RESUMO

The pharmacological effects of five benzyl - isoquinoline alkaloids were studied on the isolated perfused guinea-pig tracheal chain. The decreasing order of the spasmolytic activity exerted is: papaverine, demethoxy-papaverine, papaverinol, dioxymethverine and nitro-papaverine. The recovery of the contractile responses to carbachol depends upon the magnitude of the preceding inhibitory effect of each alkaloid and is, in some cases, incomplete. The p K IND. A values of the alkaloids, determined spectrophotometrically, are very similar and have probably no significant influence on the spasmolytic activity differences. Under the defined experimental conditions the tracheal smooth muscle shows a spontaneous development of tone that can be acellerated and increased in strength by elevating the bath temperature or by intermitent mechanical stimulation, and can be, specifically and irreversibly inhibited by indomethacin. The benzyl-isoquinoline alkaloids in small doses antagonize, reversibly, the development of tone and the contractions induced by agonist drugs.
