Estudo da adsorção de inulinases em colunas com resinas de troca ionica : paramentros experimentais e modelagem

RESUMO

The inulinases are enzymes with great commercial potencial, as they hydrolise inulin producing important products for the food industry such as fructose and fructose oligomeres. The separation and purification of these enzymes, with efficiency and compatible costs, is of great importance to the development of adsorption techniques and protocols which may optimize and amplify the operation. The objective of this work is to contribute to the improvement of the experimental techniques and mathematical modeling in the adsorption study of the inulinase enzymes on ion-exchange resins. In this work the characteristic equilibrium and adsorption kinetics parameters are determined via experiments in stirred tanks containing CM-Sepharose CL-6B and Accel Plus QMA adsorvents, allowing a good agreement between models of diffusion of sorbate on the surface and in the pores of the solid with the experimental data. Saturation experiments were made in expanded bed column with Accell Plus QMA resin and fixed bed with the two adsorvents, based on frontal analysis chromatography with a commercial mixture of inulinases-FructozymeTM. Using a complete mathematical model, where axial dispersion and resistence to mass transfer are considered, the parameters were obtained by numerical solution of the mass balance equation using the orthogonal colocation method associated with the non-linear multivariable optimization method for the obtention of the optimized mass transfer and axial dispersion coefficients. The medium values of coefficients were 0,633 1O-7m2.s-1 and 0,10 10-8 m.s-l in inulinases-CM-Sepharose CL-6B 0,747 1O-7m2.s-1 and 0,258 10-8 m.s-l in inulinases-Accell Plus QMA. The enzymes were characterised with molecular weight in 80 kDa (exo-inulinase) and 63 kDa (endo­inulinase) after gel filtration in Sephacryl S 1OO HR
