Estudo de um potyvirus isolado de especies de Crotalaria

RESUMO
A virus isolated from Grata/aria species showing the mosaic symptom, preliminarily identified as a potyvirus, was studied. The virus was named Crotalaria mosaic virus. The characteristics studied were: host range, "in vitro" properties, transmission by aphids, seed transmission and different meehanieal transmission methods, eytopathology, serologieal relation with other potyviruses and molecular mass of the capsid protein. The host range is limited to speeies of the Leguminosae family and one eharacteristie host is Phaseo/us vulgaris Preto G-2 , whieh shows local neerosis followed by systemie neerosis. The "in vitra" properties parameters were similar to the values reported for most potyviruses. Transmission of the virus by Aphis gossypíi, Myzus persicae e M. nicotíanae was demonstrated. Mechanieal transmission was effieient (80% transmission), and transmission by knife blade also was possible but with lowefficieney. Seed transmission was not observed. intracellular inelusions observed by light and electron microscopy were pinwheel and seroll type. The purifieation of the virus was possible with the proeedure used but virus yield was poor. Comparisons by PT A-ELlSA tests showed serologieal relation with the Brazilian isolate of passion-fruit woodiness virus (PWV). Using antiserum against PWV was possible to deteet erotalaria mosaie virus by PT A-ELlSA and western-blot. The relative molecular mass of the capsid pratein was estimated to be 37 kDa. Probably erotalaria mosaie virus is a strain of PWV that evolved toward a narrow host range of speeies mainly belonging to the Leguminosae family. Reeent reports indicate that most Brazilian isolates of PV N are more related to the South African Passion-fruit virus, differing from standard type PWV. This fact may explain the differences observed in the morphology of cytoplasmic inclusions from data reported for PV in other countries
