Hidrogel sintetico para reparo de defeitos da cartilagem articular

RESUMO

This work shows a study carried out with synthetic hydrogels aiming the obtention of a biomaterial to be used in the repair of articular cartilage defects. Hydrogels samples of poly (2-hydroxy ethyl methacrylate) (polyHEMA) and blends of polyHEMA with cellulose acetate (CA) or poly(methyl methacrylate-co-acrylic acid) (poly(MMA-co-AA)) was obtained by thermal polymerization and the following characterisation procedures was carried out: determination of the equilibrium water content and saline solution content, evaluation of the mechanical behaviour by creep indentation tests and cytotoxicity through in vitro tests. The results showed that it is possible to improve the mechanical behaviour of the polyHEMA hydrogels by-changing the crosslinking agent amount or by blending polyHEMA with linear polymers. The mechanical properties improve was accompanied by a decrease in the equilibrium water content and let to the incorporation of negative fixed charges to the macromolecular network, mimicking the extracelular matrix of the hyaline cartilage. This hydrogel showed characteristics similar to that one of hyaline cartilage and was used in a in vivo test that showed its potential for using in the repair of articular cartilage defects. The hydrogel implant did not show significative inflammatory reaction, let to a decrease in the synovial inflammations and improved the repair of the articular cartilage defects performed in the intercondilian region of rat femur, by forming an articular surface showing better biomechanical characteristics than the surface of biological repair in case of control defects...
