RESUMO

Clinical reports have suggested an unusual frequency in skin prik test of atopic patients using Aleuroglyphus ovatus (Ao) a storage mite as allergen. To evaluate the most important Ao antigenic fractions we submitted the Ao to different conditions of extractions. Three extractiors solutions were investigated, ammonium acid carbonate, phosphate buffer saline and bicarbonate solution (Coca solution). The soluble antigens were analyzed by SDS-PAGE, Gel filtrations, High performace chromatography (HPCL) in reverse phase:Immunobloting and skin prick test (spt. To characterize the biochemical properties of the fractions, the fractions obtained were assessed by protease?s assy. The gel filtrations (sephadex G-75) allowed a fraconation of Ao in 3 ?pools? of fractions : A, B and C. Patients test with those fraction were reactive to all of them. Nevertheless, the frequency of the reactivity od fraction C was higher than A or B. Immunoblotting showed a specific binding on the Ao fractions, molecular weights ranged from <12 to 70 kDA. Our results compared to the literature allow biochemical classification. Faction B belonging to serine protese group, Fraction C as a cystein protease. Fraction A showed mixed properties, cysteinprotease, serine and metaloprotease. Finally, to assess possible differences among allergenic sensitivity to Aleuroglyphus ovatus Blomia tropicallis and Dermatophagoides pternyssinus atopic, patients with asthmas or rinitis were tested ...Note: The complete abstract is available with the full electronic digital thesis or dissertations
