Identificação e mapeamento de genes para uso na seleção assistida de genótipos de soja com ausência do inibidor de tripsina Kunitz e lipoxigennases

RESUMO
Soybean (Glycine max (L.) Merrill) is a food with high levels of proteins, minerals and fibres, and a reduced amount of saturated fat bringing benefits to humans health. However, the use of the product for human and animal feeding is limited to the presence of some antinutritional factors, as trypsin inhibitors and lipoxygenases enzymes. The development of cultivars of soybean without these factors has been one of the objectives of some genetic improvement programs to increase the quality of food and also reduce costs in thermal processing to inactivate these components. Markers Assisted Selection (MAS) has shown to be an efficient tool in the Plant Breeding Programms to improve the efficiency selection providing more speed and praticity. The aim objectives of this work were to map the locus responsible for the expression of trypsin inhibitors in the seeds (KTi3) and the locus of Lipoxygenases 1 and 2 (Lox 1, Lox 2) using SSR markers on the F2 population BRS 213 x BRS 155 and also select plants with absence of KTi3 through the help of specific markers SCAR. The 93 plants of the F2 population had been phenotypically characterized for the presence and absence of KTi3, Lox 1, Lox 2 and Lox 3 and also genotypically characterized for KTi3 with SSR polymorphic markers Satt 409, Satt 228, Satt 429, Satt 333 and with a specific marker based on the recessive mutant allel. For Lox 1 and Lox 2 the F2 population had been characterized with the SSR markers Sat 090 and Satt 417. The markers Satt 409 and Satt 228 mapped the KTi3 gene at the A-2 soybean linking group with a distance of 17,48cM and 29,64cM respectively. For KTi3 the specific marker was 100% in efficiency. The markers Sat 090 and Satt 417 mapped the genes Lox 1 and Lox 2 at the group F of the soybean linking map with a distance of 3cM and 2,77cM respectively, being efficient in the assisted selection.
