Influencia da lectina de Crotalaria paulina e lectina do veneno de serpente Bothrops jararacussu sobre a atividade proteolitica da microbiota cariogenica

RESUMO

Lectins are polyvalent carbohydrate-binding proteins which agglutinate red blood cells. Identified in plants and animals, have also been found in snake venoms. The purpose of this study was to measure inhibition of proteolytic activity in supragingival plaque, coronal and root dentinal caries samples by CrpL e BJcuL. A standardized excavator ( 1 mg wet weight) was used to collect the samples (30 supragingival plaque; 30 - coronal caries; 30 - root caries) which were placed in 2 mL of a buffering solution (pH 7,2). The samples were vortexed for 30 seconds and 250 ?L were added to: A) 250 ?L CrpL 0,12% + 250 ?L BANA 1.67 mmol/L + 250 ?L distilled water; B)250 ?L BJcuL 0,12% + 250 ?L BANA 1.67 mmol/L + 250 ?L distilled water; C) 250 ?L CrpL 0,12% + 250 ?L BANA 1.67 mmol/L + 250 ?L N-acetil-D-galactosamine 25 mM; D) 250 ?L BJcuL 0,12% + 250 ?L BANA 1.67 mmol/L + 250 ?L lactose 0.8 mM. The mixtures were incubated for 18 hours at 37°C and color was developed by the addition of 50 ?L of 0,1% Fast Garnet. The absorbance at 510 nm was recorded spectrophotometrically. Independent t-tests were employed to determine differences between mean optical densities of the experimental groups and controls.The mean absorbance of CrpL group [0.375 (±0.028)] and BJcuL group [0.442 (±0.057)] was significantly different than 0.860 (±0.056) and 0.753 (±0.058) control groups (p <0.001) in supragingival plaque samples. The mean absorbance of CrpL group [0.471 (±0.044)] and BJcuL group [0.563 (±0.035)] was significantly different than 0.860 (±0.056) and 0.753 (±0.058) control group (p <0.001) in coronal caries. The mean optical density of CrpL group [0.684 (±0.027)] and BJcuL [0.789 (±0.046)] wasn t significantly different than 0.742 (±0.030) and 0.753 (±0.058) control groups. It is concluded that CrpL and BJcuL can inhibit proteolytic activity present in human supragingival plaque and coronal caries
