RESUMO

ln this work, we investigated the effeets of MAPA on the immune response. We have stlldied the neutrophils exposed to these eompound of six normal individuaIs. Phagoeytosis and killing of Candida albicans and Candida ke.fyr by neutrophils, neutrophil ehemotaxis and nitroblue tetrazolium reduetion were measured. The results obtained demonstrated - an inerease in the phagoeytie funetion of neutrophils in exposed to MAPA. On the other hand, lysis of C. albicans and C. kefyr by neutrophils was signifieantly inereased. We also observed a signifieant inerease of nitroblue tetrazolium reduetion and neutrophil ehemotaxis in these eells. e also investigated the effeets of magnesium ammonium phospholinoleatepalmitoleate anhidryde (MAPA), a roteie aggregated polymer isoleted from Aspergillus orizae, on the growth and differentiation of granuloeyte-maerophage progenitor eells (CFU-GM) in normal and Listeria monocytogenes infeeted miee. MAPA was administred subeutaneously (0.5-7.5 mg/Kg) for 7 days. ln the MAPA treated/infeeted groups L. monocytogenes was administred at the end of MAP A treatment. A signifieant reduetion in the numbers ofCFU-GM was observed at 48 h after the infection, followed by a slight restoration at 72 h. Treatment of, these animaIs previously to infection stimulated marrow myelopoiesis in a dosedependent manner. The 5.0 mg/Kg MAPA was the optimal biological aetive dose in infected mice and this dose shedule also stimulated myelopoiesis in normal mice. The 0.5 and 5. O mg/Kg doses given prior to in.fection with a lethal dose of 3 x 108 bacterialanimal led to a 30% and 40% survival, respectively. These results demonstrated that MAPA produces a significant increase in the resistanee of the animaIs infeeted with L. monocytogenes, and that this protection is due, at least in part to increased CFU-GM in the bone marrow ofinfeeted animals
