Obtenção da enzima xilose redutase de Candida mogii e sua purificação em sistemas de duas fases aquosas

RESUMO

The obtainment, extraction and partial characterization of the enzyme xylose reductase (XR) present in cell extract of the yeast Candida mogii NRRL Y-17032 cultivated in rice straw hemicellulosic hydrolysate were studied in this work. The XR production was evaluated by varying the concentration of hydrolysate in the growth medium, and the highest values of production and specific productivity (35 U/g of cell and 0.97 U/[g of cell.h], respectively) were obtained in fermentations employing medium containing 49.2 9 xylose/l. The conditions for in vitro optimal XR activity were studied and the maximum volumetric activity (0.54 U/ml) was obtained employing pH 6.5 and 38°C. In the study of the stability, XR activity remained constant for 3 hours at temperatures of 4 and 38°C, and for 4 months at -18°C. The kinetic parameters determined for XR were Vmax = 0.485 U/ml and Km = 63 mM for xylose and Vmax = 0.390 U/ml and Km = 0.032 mM for the cofactor NADPH. For the disruption of C. mogii cells by mechanical agitation with glass beads, the highest volumetric activity (0.683 U/ml) was found employing 300 ?m diameter beads, cell concentration of 45 g/l and 50 cycles of agitation. In the study for XR extraction in aqueous two-phase systems, mathematical models were predicted for the purification factor and percent recovery (yield) on top phase, and the maximum values predicted by these models were 1.59 and 105.8, respectively
