RESUMO

Hycanthone is a drug used for the clinical trials of schistossomiasis, although inducing chro mosome breaks when it enters the DNA double helix . In this work the binding of hycanthone to chromatin areas dif fering in terms of composition and phisyology, was inves tigated. Based on visual observation of nu clear basophilia and anisotropy a preferential binding of hycanthone to heterochromatin could be demonstratatin the nuclei of the Malpighian tubules of T. in6e_tan_ Klug. However, in other cellular systems ( mouse lymphocytes, Kid ney cells in culture and plant cells ), the drug can also bind to euchromatin irrespective of its compactness leveI. When penetrating the various hete rochromatin types ( except in the case of T. in6e_tan_ he terochromatin ), the drug induces chromatin loosening,that certainly favors incidence of chromatin breaks. The varia tion of hycanthone b_nding to the DNA in the cell types studied in this wo_k are possibly related to differences in composition, arrangement and stability of DNA-protein com plexes involved.
