RESUMO

Plasma from a patient (JJO) with multiple myeloma showing high levels of M paraprotein was obtained from the faculdade de Medicina, Universidade Estadual de Campinas.The results of electrophoretic analysis followed by densitometry showed a compact band indicative of molecular homogeneity characteristic of patient presenting monoclonal gamopathy. Fraction A containing myeloma protein was isolated from the pathologyc serum JJO by ion exchange chomatografy in DEAE celulose. Electrophoretic control of this fraction indicates the purification of the M component, without contamination with other plasma proteins. Fraction A was typed serologicaly as an IgGl immunoglobulin with lambda light chains. The purity of JJO protein (fraction A) was confirmed in SDS-polyacrylamide gel electroforesis. The reduced JJO protein showed two bands of 50 and 25K corresponding to heavy and light chains respectively. The M (JJO) paraprotein in was chemically typed as an IgGl protein with lambda light chains by autoradiography of high voltage electrophoresis of a pepsin-trypsin digests of myeloma protein partially reduced and alkylated with iodoacetate-C14. This material produces a consistent autoradiographic pattern. Which shows radioactives bands Jl, J2 andJ3 of electrophoretic mobility characteristic the H-L, H-H and L. peptides respectively
