Pressão hidrostática : efeito na verificação, ultraestrutura e expressão gênica de embriões bovinos

RESUMO

Despite the enormous growth achieved by the technique of bovine embryo in vitro production in recent years, a limitation is still evident, these embryos are more susceptible to cryopreservation than their counterparts in vivo produced. To obtain better results in the cryopreservation procedure, researchers are seeking improve embryos in vitro produced quality or change cryopreservation systems. The aim of this work was to define parameters for the use of hydrostatic pressure as a new tool in the process of vitrification of bovine embryos produced in vitro by evaluating the effects of it on ultrastructurally and gene expression on these embryos. The results indicated that embryo pressurization at 80 MPa had become deleterious to embryonic development. But pressure level at 60 MPa for 1 hour improved hatching rate after embryo vitrification compared to only vitrified group (79% vs 66%). And 1 hour after pressurization would be the best time to vitrification, with higher rates of re-expansion, hatching (90% and 74% respectively), increased speed of re-expansion after vitrification/devitrification and higher abundance of ERG 25 transcripts at this time, compared with the control group, immediately and 2 hours after the pressure. Ultrastructurally, embryos submitted to pressure had greater amount of microvilli and increased mitochondria hooded that control embryos. In the group of vitrified embryos after pressure there were a greater number of nuclei per area when compared to the only vitrified group. Certain levels of hydrostatic pressure treatment improved the in vitro survival vitrified bovine embryos. Embryonic cells react in response to stress, increasing the expression of specific genes, such as ERG 25, and changing ultrastructure, which may promote increased resistance to other stressful procedures, such as vitrification.
