RESUMO

Lectins, proteins that recognize carbohydrates specifically and reversibly, can be purified by affinity chromatography. The genus Bauhinia (Fabaceae) includes native or introduced species that have been used in the treatment of diabetes. A galactose-specific leaf lectin (BmoLL) has been previously purified from B. monandra and available in milligram quantities; rabbit anti-BmoLL IgG was also available, as such and conjugated to peroxidase. The aim of this investigation was to produce lectins in different tissue cultures from B. monandra (callus, conventional and Agrobacterium rhizogenes-transformed roots), immunodetect lectin in different B. monandra preparations through double immunodiffusion assays and electrophoretic transfer techniques, as well as to evaluate leaf extracts of the plant as a source of antioxidant and hypoglicemiant compounds. Cultures were obtained from B. monandra seeds; extracts (10 %, w/v) and F 0-60% preparations were performed with 10 mM citrate phosphate buffer, pH 6.5. Haemagglutinating activity was detected in F 0-60% of different tissue cultures, when tested with erythrocytes from rabbit and human blood group AB. Immunodiffusion with anti-BmoLL IgG revealed precipitation lines in all cultures. SDS-PAGE of F 0-60 % from different tissue cultures showed a band pattern of molecular weights similar to BmoLL. Polypeptides of different preparations from B. monandra interacted with anti-BmoLL IgG. B. monandra leaves contain at least three different compounds with antioxidant activity. It was investigated the acute effect of an ethanolic extract from B. monandra leaves (E) on glucose and insulin levels of control (CE) and intrauterine malnutrition (ME) Wistar rats in comparison with animals submitted to vehicle (V) and groups CV and MV. Acute E administration reduced the glucose level, but did not change the secretion of insulin in C rats. The capacity of rats submitted to intra-uterine malnutrition to secrete insulin was apparently restored; hypoglycemia induced by E was due to insulin secretion
