RESUMO

The aim of this work is to evaluate xanthan production in a bubble column reactor using a repeated batch process as well as to optimize de gum production operational conditions and evaluate some of the typical steps of the recovery process. Two Xanthomonas campestris strains were evaluated: ATCC 13951 (or NRRL B-1459) and ATCC 55298. The cycles were comprised of 3 to 8 repeated 48-hour batches. At the end of each batch 20% of the broth volume was left inside the fermentor to serve as innoculum to the next bacth that were initiated by addition of fresh medium. Preliminary experimental assay showed satisfactory results in a cycle with six 48-hour successive batches so that subsequent assays were conducted to optimize the operational conditions for each strain using two level factorial design plus star configuration with three central point replicates. The factors evaluated were the initial sucrose concentration, temperature and aeration and the responses were the final xanthan concentration, the overall yield and the consistency index. The evaluated factors did not presented significant effect on the responses studied. The mean values obtained in the assays with strain ATCC 13951 were 10.7±1.9 g/L to final xanthan concentration, 48.6±14.3% to the overall yield, 1228±310 mPa.sn to the consistency index and 0.29±0.022 to the flow behavior index. The mean values obtained in the assays with strain ATCC 55298 were 11.1±1.6 g/L to final xanthan concentration, 50.5±11.7% to the overall yield, 1478±223 mPa.sn to the consistency index and 0.27±0.016 to the flow behavior index. The analysis of the factorial design results through desirability function pointed the optimal conditions to maximize yield and product quality and these conditions were validated experimentally. It was concluded that for both strains, the optimal conditions were 20g/L initial sucrose, 31ºC and 1.5 vvm.
