Recuperação das enzimas Alfa e Beta - amilases em sistema bifasico aquoso PEG / CACL2 para uso como biocatalisador de amilaceos

RESUMO

Aqueous two-phase systems (A TPS) PEG/ CaCl2 was studying in pH 5,0; 6,0 and 7,0 and polymer molecular weight 4000, 6000 and 8000, in environment pressure and temperature. The enzymes ? and ?-amylases from maize malt was recovered in bath and continuous extraction process, it was valorized this product. The enzymes were obtained by germination of maize seeds. It was determinate of the enzymatic activity by modify Wohlghnuth method, Sandstedt, Kneen, Blich (1959) and Milles Laboratory method (1958), showed in REGUL Y (1996), and protein total by BRADFORD method (1976). The kinetic study was making between the enzymatic extract in PEG and maize malt, in optimum condition of enzymes. The enzymes were recovered in bath and continuous process systems. It was making an experimental plain for process optimization. A TPS PEG/ CaCl2 had similar conduct to literature description. The adjusted empiric model to bath partition was linear with lnK. The combination of partition coefficient with enzymatic activity optimums showed the best system for recovery of the enzymes as been the A TPS PEG 4000/ CaCl2, at pH 5,0. The kinetic study introduced a high catalytic power in enzymatic extract in PEG. The optimization of the continuous extraction process carried to a quadratic model of the pallet rotation and flow rate with the partition coefficient, where optimum region showed it in the value minor of the independents variables
