1. Quantificação de forças na manipulação de nano-objetos individuais em experimentos "in situ" de mocroscopia eletronica / Quantification of forces on the manipulation of individual nano-objects in "in situ" experiments of electron microscopy

2. Nanomanipulation and characterization of individual nano-objects for in situ experiments of electron microscopy / Nanomanipulação e caracterização de nano-objetos individuais por experimentos in situ de microscopia eletronica
It is expected that, in the future, high-technology devices should be based on new and unexpected physical and chemical properties of nanometric objects. Many applications require nano-objects to be selectively positioned at well-defined positions of a device. However, the well-established methods of physical manipulation used in the macroscopic scale are no

3. Direct measurement of stiffness of single actin filaments with and without tropomyosin by in vitro nanomanipulation.
In order to explain the molecular mechanism of muscle contraction, it is crucial to know the distribution of the sarcomere compliance of active muscle. Here, we directly measure the stiffness of single actin filaments with and without tropomyosin, using a recently developed technique for nanomanipulation of single actin filaments with microneedles. The resul

4. Modelling extreme stretching of DNA.
Molecular modelling with Jumna is used to study extreme stretching of the DNA double helix. The results, which correlate well with recent nanomanipulation experiments, show how the double helix can be extended to twice its normal length before its base pairs break. Depending on the way the duplex is stretched two types of conformation can occur, either an un

5. Multimodal optical workstation for simultaneous linear, nonlinear microscopy and nanomanipulation: Upgrading a commercial confocal inverted microscope
In this work we propose and build a multimodal optical workstation that extends a commercially available confocal microscope (Nikon Confocal C1-Si) to include nonlinear∕multiphoton microscopy and optical manipulation∕stimulation tools such as nanosurgery. The setup allows both subsystems (confocal and nonlinear) to work independently and simultaneously.

6. Promoter unwinding and promoter clearance by RNA polymerase: Detection by single-molecule DNA nanomanipulation
By monitoring the end-to-end extension of a mechanically stretched, supercoiled, single DNA molecule, we have been able directly to observe the change in extension associated with unwinding of approximately one turn of promoter DNA by RNA polymerase (RNAP). By performing parallel experiments with negatively and positively supercoiled DNA, we have been able t
