1. SELEÇÃO POLIFÁSICA DE MICRORGANISMOS PRODUTORES DE POLIHIDROXIALCANOATOS
Polyhydroxyalkanoates (PHAs) are fully biodegradable biopolyesters produced by many prokaryotes and accumulated as insoluble cytoplasmatic inclusions. The detection of these intracellular granule is usually provided using lipofilic azodyes, which are not specific. Another way to screen PHA-producing bacteria is through culture-independent molecular technique

2. Separation of polyhydroxyalkanoates-producing bacterial strains using PHA synthase gene and their evaluation for PHA deposition
In this study, a variety of samples were screened for the presence of PHA synthase gene. Results showed that 16 out of 102 isolated were positive for PHA respective genes. The highest prevalence was observed in Pseudomonas aeruginosa. The capability of PHA production was also shown by growing these strains on the defined medium and subsequent analysis using

3. Phytohemagglutinin improves the development and ultrastructure of in vitro-cultured goat (Capra hircus) preantral follicles
The objective this study was to determine the effect of phytohemagglutinin (PHA) on survival, growth and gene expression in caprine secondary follicles culturedin vitro. Secondary follicles (∼0.2 mm) were isolated from the cortex of caprine ovaries and cultured individually for 6 days in α-MEM+ supplemented with PHA (0, 1, 10, 50, 100, or 200 µg/mL). Aft

4. Isolamento de bactérias produtoras de polihidroxialcanoatos e caracterização molecular de sua PHA sintase. / Isolation of polyhydroxyalkanoates producing bacteria and molecular characterization of their PHA synthase.
A total of 1520 isolates from domestic sewage sludge or soil of preserved area were evaluated in regards of PHA production using the lipophilic Nile Red A or Sudan Black B dyes, allowing the detection of 261 isolates. PHA production experiments revealed P3HB producing isolates at higher amounts than the strain Burkholderia sacchari LFM101 and PHAMCL producti

5. Avaliação do potencial de Burkholderia sacchari produzir o copolimero biodegradável poli(3-hidroxibutirato-co-3-hidroxihexanoato) [P(3HB-co-3HHX)]. / Evaluating the potential of Burkholderia sacchari to produce the biodegradable copolymer poly (3-hydroxybutirate-co-3-hydroxyhexanoate).
The ability of B. sacchari to accumulate poly-3-hydroxybutyrate-co-3-hydroxyhexanoate (P3HB-co-3HHx) from glucose and hexanoic acid was confirmed. 3HHx content was up to 2 mol% of PHA (<10% of the maximum theoretical 3HHx from the acid), indicating a substrate flexibility of B. sacchari PHA synthase, but high efficiency of hexanoate catabolic pathways. 6. Obtenção de mutantes deficientes no acúmulo de PHA e construção de linhagens recombinantes para o controle da composição monomérica / Mutants deficient on polyhydroxyalkanoate (PHA) accumulation and construction of recombinants to control monomer composition on PHA

Identification of genes involved in the synthesis of polyhydroxyalkanoates on Burkholderia cepacia strain IPT64.

9. The Composition a evaluate the polymer biodegradable produce for Burkholderia sacchari with the different substrates / Avaliação da composição de polímeros biodegradáveis produzidos por Burkholderia sacchari a partir de diferentes substratos
The bacterial production of polyesters belonging to the polyhydroxyalkanoate family (PHA) has generated biodegradable, biocompatible and environmentally friendly new materials. Properties and applications of PHA depend on monomer composition, which is dependent both on the substrate available and the metabolic apparatus of the PHA-producing microorganism. Bu

10. PhaC and PhaR Are Required for Polyhydroxyalkanoic Acid Synthase Activity in Bacillus megaterium
Polyhydroxyalkanoic acids (PHAs) are a class of polyesters stored in inclusion bodies and found in many bacteria and in some archaea. The terminal step in the synthesis of PHA is catalyzed by PHA synthase. Genes encoding this enzyme have been cloned, and the primary sequence of the protein, PhaC, is deduced from the nucleotide sequences of more than 30 organ

11. Cloning, Molecular Analysis, and Expression of the Polyhydroxyalkanoic Acid Synthase (phaC) Gene from Chromobacterium violaceum
The polyhydroxyalkanoic acid synthase gene from Chromobacterium violaceum (phaCCv) was cloned and characterized. A 6.3-kb BamHI fragment was found to contain both phaCCv and the polyhydroxyalkanoic acid (PHA)-specific 3-ketothiolase (phaACv). Escherichia coli strains harboring this fragment produced significant levels of PHA synthase and 3-ketothiolase, as j

12. Genetic Analysis of Comamonas acidovorans Polyhydroxyalkanoate Synthase and Factors Affecting the Incorporation of 4-Hydroxybutyrate Monomer
The polyhydroxyalkanoate (PHA) synthase gene of Comamonas acidovorans DS-17 (phaCCa) was cloned by using the synthase gene of Alcaligenes eutrophus as a heterologous hybridization probe. Complete sequencing of a 4.0-kbp SmaI-HindIII (SH40) subfragment revealed the presence of a 1,893-bp PHA synthase coding region which was followed by a 1,182-bp β-ketothiol
